Mouse | MMP20 | 9313 | matrix metallopeptidase 20 | Expression of collagen XVIII and MMP-20 in developing teeth and odontogenic tumors. In odontogenic tumors MMP-20 and collagen XVIII were co-localized in the enamel-like tumor matrix. | Human | COL18A1 | 80781 | collagen, type XVIII, alpha 1 | Collagen XVIII and enamelysin were co-localized in the developing enamel matrix and stratum intermedium and in the enamel-like tumor matrix of odontogenic tumors | Human | ODAM | 54959 | odontogenic, ameloblast asssociated | Calcifying epithelial odontogenic tumor-associated amyloid consists of a unique and previously undescribed protein that we provisionally designate APin | Human | ENAM | 10117 | enamelin | In view of the present results, it is interesting that previous studies have indicated that although ameloblastoma, a non-mineralized odontogenic tumor, transcribes amelogenin mRNA, amelogenin (and enamelin) proteins are not expressed in this tissue. Immunohistochemical localization of two enamel proteins, amelogenin and enamelin, in comparison with that of keratin, was determined in odontogenic tumors and the allied lesions in order to verify functional differentiation of the tumor cells as ameloblasts. Amelogenin and enamelin were demonstrated in small mineralized foci and in the tumor cells surrounding them in adenomatoid odontogenic tumor (AOT), calcifying epithelial odontogenic tumor (CEOT), and calcifying odontogenic cyst (COC). | Human | MMP20 | 9313 | matrix metallopeptidase 20 | Thus, taken together, our results suggest that the enamelysin-specific monoclonal antibody (203-1C7) may be utilized as a marker of early enamel development and that enamelysin may be involved in the pathogenesis of specific odontogenic tumors. Human matrix metalloproteinase-20 (MMP-20, enamelysin) fragments the enamel-specific protein amelogenin and has been shown to be synthesized exclusively by odontoblasts and ameloblasts and in certain odontogenic tumors. Since this protease is thought to play important roles in tooth development, the evaluation of enamelysin in odontogenic tumors may aid our understanding of the histogenesis and cell differentiation of such lesions. Furthermore, enamelysin was also detected in globular amyloid masses and calcified foci in calcifying epithelial odontogenic tumors, hyaline droplets, small and large mineralized areas in adenomatoid odontogenic tumors, and a portion of ghost cells in calcifying odontogenic cysts. Immunohistochemical detection and distribution of enamelysin (MMP-20) in human odontogenic tumors. BACKGROUND: To clarify the roles of Sonic hedgehog (SHH) signal transduction in oncogenesis and cytodifferentiation of odontogenic tumors, expression of SHH, Patched (PTC), Smoothened (SMO), and GLI1 was analyzed in ameloblastomas as well as in tooth germs. Odontogenic tumor tissues including 34 epithelial derived, 24 epithelial-mesenchymal derived, and 26 mesenchymal derived were examined by immunohistochemistry for SHH, PTC, SMO and GLI1. The findings suggest SHH, PTC, SMO and GLI1 protein are predominantly located in epithelial components in various odontogenic tumors and might participate in the proliferation of epithelial components of odontogenic tumors. CONCLUSION: Expression of SHH, PTC, SMO, and GLI1 in tooth germs and ameloblastomas suggests that these SHH signaling molecules might play a role in epithelial-mesenchymal interactions and cell proliferation in tooth development as well as in growth of these epithelial odontogenic tumors. Immunoreactivity for SHH, PTC, SMO and GLI1 was detected in both epithelial derived odontogenic tumors and epithelial-mesenchymal derived odontogenic tumors with or without dental hard tissue formation. | Human | SHH | 6469 | sonic hedgehog | CONCLUSION: Expression of SHH, PTC, SMO, and GLI1 in tooth germs and ameloblastomas suggests that these SHH signaling molecules might play a role in epithelial-mesenchymal interactions and cell proliferation in tooth development as well as in growth of these epithelial odontogenic tumors.
